The work of the Greco-Roman physician Galen was considered the true and most important source of medical knowledge for more than fifteen centuries. Galen considered that nerves are connected to the brain, either directly or through the spinal cord. He performed experiments on animals by cutting the spinal cord at various heights and observed the loss of control of various functions of the animal. He correctly predicted the effect of cutting the phrenic nerve, which controls the diaphragm in breathing; he checked the compass value of pulsations, and the response to emotional stresses; he demonstrated that the kidneys process urine and showed that arteries contain blood, not water.

Claudius Galen of Pergamos (129-216) was, together with Hippocrates, one of the greatest physicians of the classical world and his influence dominated medical knowledge until the 18th Century. He was the author of a vast work written in Greek, which has come down to our days, partly in the original Greek and partly in ancient Latin translations. It is an encyclopaedic summary which is not limited to the various branches of medicine but which studies it in context with knowledge of other áreas, such as philosophy, mathematics, architecture, philology and literature. During the Middle Ages he was less known in Europe, but his works continued to be read and disseminated in Byzantium and translated by Arabic physicians. The greater knowledge of Galen's work from the Renaissance onwards boosted the development of medicine, as the Greek physician attached great importance to experimentation and the study of anatomy.

The most important edition of this work, printed by Aldo Manucio in 1525 (vide Brunet Vol II col. 1148) culminated the scientific collection of Galen's works that had lasted since the 15th century, marking a new era in medical education and in medical philology and terminology.

At the beginning of the 16th century, Galen's works, like those of other classical authors, were scattered in numerous manuscripts, each of which contained only a fragment. The 16th century collection was carried out by the professors of Italian universities, namely the University of Pavia. G. B. Opizzoni (ca. 1485-ca. 1532), coordinated a group of English physicians from northern Italy: John Clement (ca. 1495-1572), Edward Wotton (1492-1555), William Rose (ca. 1490-1525), Thomas Lupset (1495-1530), all disciples of Thomas Linacre (ca. 1460-1524), and the Saxon George Agricola (1495-1555).

The aldin princeps edition of all Galen's works in Greek, printed in 1525, was possible due to the expertise and understanding that the Manucio printers had of the Greek language. Until then there were only Latin editions of parts of Galen's works published in Venice (1490) and Pavia (1502, 1511, 1515/1516, 1522, 1528), and so on. The Venetian Latin edition of the Iuntas subsequently served for the Leonese editions from 1550 onwards. The Latin editions were initially based on Galen's treatises written in Arabic. However they were successively compared with the Greek language editions and added with commentaries by the most famous physicians of the late four hundred and early five hundred, such as Vesalius. This ninth edition of the Iuntas presents the monumental systematic and remissive index of Brasavoli, which occupies the entire last volume, in addition to many other indexes by other authors, already present in this and previous editions.
